He said the blockade was "impairing United States and other global business activities in the region" and that the USA backed mediation efforts being pursued by Kuwait.
He said Qatar remained committed to a Kuwaiti-led mediation effort, but that he had yet to receive a clear list of demands.
"Fighting terrorism and extremism is no longer a choice, rather ... a commitment requiring decisive and swift action to cut off all funding sources for terrorism regardless of its financier", the official Saudi Press Agency reported, citing a government source.
On Saturday, Saudi Arabia, Bahrain and the UAE made no mention of Mr Tillerson's call.
UAE ambassador to the US Yousef al-Otaiba praised Mr Trump's leadership in the face of Qatar's "troubling support for extremism".

He said that up to 11,000 foreign militants had allegedly arrived in Afghanistan in the last two years to fight for Daesh. Afghan authorities have said Pakistan was involved in the attack, charges denied by Islamabad.
The incident triggered a diplomatic row between Qatar and its neighbors.
Al-Thani, during a visit to Russian Federation to discuss the crisis, said: "The US sees Hamas as a terror organization, but to the rest of the Arab nations it is a legitimate resistance movement".
Saudi Arabia said an immediate change of policy by Qatar was essential.
Russia's top diplomat Sergey Lavrov and Qatari Foreign Minister Sheikh Mohammed bin Abdulrahman Al Thani met in Moscow Saturday.
External affairs ministry said India is closely following the emerging situation in the region in the wake of the recent decision by Saudi Arabia and some other countries to break diplomatic relations with the State of Qatar. "However, we can not be pleased with the situation, where these relations between our counterparts are aggravating".
On Tuesday, Saudi Foreign Minister Adel al-Jubeir said that to rebuild relations, Doha must cut its support for "extremist" groups, including Hamas.
"The nation of Qatar, unfortunately, has historically been a funder of terrorism at a very high level", Trump said, echoing an allegation the Saudi-led group has used to justify cutting diplomatic ties to the tiny gas-rich country.
